Description

Movie shows spindle assembly around DNA-beads in a Xenopus laevis egg extract. DNA beads are autofluorescent, and spindle is formed from exogenous X-rhodamine-labeled tubulin and alexa-488 labeled anti gamma tubulin antibody. Upper left panel shows the alexa-488 signal, upper right the X-rhodamine signal, and lower left the merge (red and green respectively). By the end of the movie a well-defined spindle has formed with the poles marked by the green gamma tubulin signal. DNA beads are autofluorescent in red and green channels. Images were capured every 60 sec with a Hamamatsu Orca-ER cooled CCD camera using a Nikon TE2000-U stand and a 40x 1.3 NA objective lens in widefield mode.
