A High resolution micrograph of tetrahymena showing a transverse ribbon starting from the anterior right side of the proximal end of a basal body (out of section plane) and crossing under the pellicle to end under the longitudinal ribbon in the adjacent ridge crest. The epiplasm separates the two ribbons. Mitochondria often lie in ridges and the crescent end of kinetodesmal fiber, a dictyosome, and the endosomal membrane is seen. TEM taken on 8/25/67 by R. Allen with Philips 200 operating at 60kV. Neg. 19,200X. Bar = 0.2µm. The raw film was scanned with an Epson Perfection V750 Pro. This image is best used for quantitative analysis.
Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. Additional information available at (http://www5.pbrc.hawaii.edu/allen/).
