Description

Cytosolic pressure may be one force used to expel the undigested contents from spent vacuoles, however, tubulation and retrieval of the spent vacuole membrane will ultimately eliminate all of the vacuole contents as well as the spent vacuole membrane and allow the cytoproct lips to be pulled back together. The vesiculated vacuole membrane can then be recycled. TEM taken on 4/21/78 by R. Allen with Hitachi HU11A operating at 75kV. Neg. 12,250X. Bar = 0.5µm. The negative was printed to paper and the image was scanned to Photoshop. This digitized image is available for qualitative analysis. Technical Details

Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. Additional information available at (http://www5.pbrc.hawaii.edu/allen/).
