Isolated chloroplas tmembranes of the unicellular photosynthetic alga Chlamydomonas reinhardtii were fixed, sectioned, and examined by transmission electron microscopy. In this ac-5 mutant, regions of stacked and unstacked thylakoids are seen. Isolated chloroplast membranes were fixed with glutaraldehyde, post fixed with osmium tetroxide, dehydrated, embedded in epoxy resin, and thin sections cut, stained and examined with a Hitachi HU-11C TEM.
